摘要:
Embryo development and disease progression is actually a journey of gradual acquisition of specific functions with the continuous specialization of cell fate, accompanied by the accumulation of cell heterogeneity within the embryo and adult. Here, with the single-cell in vitro/vivo functional and omics analysis, we will talk about the lineage specification (e.g. endothelial-to-hematopoiesis transition in hematopoietic stem cell emergence) and the pluripotency transition (e.g. naive-to-primed transition in implanting epiblast) during early mammalian/human embryo development. Understanding the multi-dimensional molecular mechanism (e.g. gene networks coordination with epigenetic factors) of cell fate decision will not only shed light on how orderly embryo body plan from a single-cell fertilized-egg to a complex and highly functional organism, but also largely inspire what potential regulators cause the diseases evolution such as cancer, essentially a disordered transformation of cell fates.
